    No not black rot. It looks like it could be damage from feeding small insects, not sure which ones but black rot spots would be on the upper leaf and be larger and brown, not black.

    It looks similar to ladybug egg masses but they seem darker. I think I would worry more about the bad case of mildew the leaf has. Set this one aside and see what hatches take a picture and we may ID it from that.

    Both 2,4-D and dicamba are very bad for grapes. Pretty weed free lawns don't co-exist well with grapes because of this. If you keep a good thick grass lawn it will help keep the weeds out and just realize you may have to allow a few weeds to grow to be able to keep the grapes.

    Vines are resilient and can adapt to their situations. You may find that the shoots will form plenty of summer laterals with fresh leaves on them and unless those get infested, they can ripen the crop. Good luck with them.
